Discovery of Homobivalent Bitopic Ligands of the Cannabinoid CB2 Receptor**

Single chemical entities with potential to simultaneously interact with two binding sites are emerging strategies in medicinal chemistry. We have designed, synthesized and functionally characterized the first bitopic ligands for the CB₂ receptor. These compounds selectively target CB₂ versus CB₁ receptors. Their binding mode was studied by molecular dynamic simulations and site-directed mutagenesis.     

Nanotube molecular transporters: internalization of carbon nanotube-protein conjugates into Mammalian cells

The interactions between various functionalized carbon nanotubes and several types of human cancer cells are explored. We have prepared modified nanotubes and have shown that these can be derivatized in a way that enables attachment of small molecules and of proteins, the latter through a novel noncovalent association. The functionalized carbon nanotubes enter nonadherent human cancer cells as well as adherent cell lines (CHO and 3T3) and by themselves are not toxic. While the fluoresceinated protein streptavidin (MW approximately 60 kD) by itself does not enter cells, it readily enters cells when complexed to a nanotube-biotin transporter and exhibits dose-dependent cytotoxicity. The uptake pathway is consistent with adsorption-mediated endocytosis. The use of carbon nanotubes as molecular transporters could be exploited for various cargos. The biocompatibility and unique physical, electrical, optical, and mechanical properties of nanotubes provide the basis for new classes of materials for drug, protein, and gene delivery applications.     

Ultrasensitive ATP detection using firefly luciferase entrapped in sugar-modified sol-gel-derived silica

Firefly luciferase (FL) was entrapped in sol-gel-derived silica containing precursors based on covalent linkage of d-gluconolactone or d-maltonolactone to (aminopropyl)triethoxysilane to form N-(3-triethoxysilylpropyl)gluconamide or N-(3-triethoxysilylpropyl)maltonamide. The enzyme was active and stable in this material and showed catalytic constants close to those in solution. As little as 20 amol ATP could be detected with the entrapped FL, and the entrapped enzyme could be used over several cycles.     

Abstract Hardy-Sobolev spaces and interpolation

The purpose of this work is to describe an abstract theory of Hardy-Sobolev spaces on doubling Riemannian manifolds via an atomic decomposition. We study the real interpolation of these spaces with Sobolev spaces and finally give applications to Riesz inequalities.

A Study of the Phase Transition in the System [Ag1−x(Na)x]NO3 by Infrared Spectroscopy

A complete infrared study of the mixed crystal system of AgNO₃ and NaNO₃ is carried out in the region 400–4000 cm^?1. The study includes internal fundamental normal vibrations of the NO^?₃ ion in the ordered and disordered phases of AgNO₃ at different values of the sodium concentration, the I.R. spectra, spectral band shape intensities, and frequencies of the internal modes as functions of the sodium ion concentration. Special attention is paid to bending mode, combination mode, asymmetric stretching mode, and over-tone. The rotational energy barrier is determined at different concentrations of the sodium ions in the system.
